Abstract

Antibody-based diagnostics and therapeutics require biophysical characteristics in specificity, affinity, and stability. Phage-displayed recombinant antibody libraries have been powerful tools in discovering and enhancing the desired biophysical properties. However, the sequence space of the complementarity determining regions in antibody are often too vast to be explored randomly with current antibody libraries. Consequently, with rapidly expanding sequence and structural database in immunoglobulin, computational structural bioinformatics tools are becoming increasingly useful in analyzing antibody-antigen interaction and antibody stability, and more importantly, in designing synthetic antibody libraries for antibody discovery. The integration of the antibody display platform with the increasingly powerful bioinformatics in immunoglobulin is to bring further innovations in engineering antibody-based diagnostics and therapeutics.
