Abstract

In recent years, massively parallel RNA sequencing (RNA-seq) is rapidly becoming an attractive alternative to microarrays for disease association studies. RNA-seq has been widely used for identifying differential expression genes and splicing isoforms between treatment and control groups. However, existing methods often have little or no power for identifying risk genes with aberrant profiles of allele-specific expression (ASE) and isoform switching in time-series studies. In this talk, we first present our RNA-seq analysis pipeline for detecting aberrant ASE in association studies, which is useful for recessive disorders within nuclear families. Finally, we present current progress and challenges of mining isoform switching from time-series data.
